First thing came to my mind (on reading the scenario) was Rakko's "Borderline" – and perhaps there are others to which reference could be made.
Rather similar – cute little sister decides to get together with her "onii-chan", and, while he might hold off for a while, the (ahem) outcome is a foregone conclusion.

This is based on 2 episodes of Nisemonogatari, the direct sequel/second sesaon of Bakemonogatari. What you should know? Bake- and Nise- tell the story of Koyomi Araragi, the young man in this doujin. He is a former vampire and gets involved with assorted appararitions/possessions/spirits that are affecting a number of girls he knows or gets to know. The young blond girl, Shinobu? She's the remnants of the 500-year old vampire who turned Koyomi into a vampire, and their lives are linked. She lives in his shadow now. She came out and took a bath with him, chatting up for the first time in the series and they were seen when youngest sister, Tsukihi saw them together. She returned with a butcher knife, and was able to be convinced that it was her imagination of seeing Shinobu (who had gone back in Koyomi's shadow). As a former vampire, Koyomi can recover fully, and quickly from drastic wounds.
The other episode had him betting with the older of his younger sisters that she couldn't take 5 minutes of what turned out to be toothbrushing (she wanted to be introduced to an athletic girl who's a star athelete; Koyomi, however, knows that his female friend is a lesbian who likes girls younger than herself, and didn't want to expose his sister to that friend's tendencies).
So, it's inspired as a mish-mash of two episodes, with hentai thrown in.
